I've seen this effect used frequently so I'm hoping that it's fairly easy!
I'm just trying to set an image so that it's one color and have certain parts of it transparent so that it blends into the background and becomes two-toned.

Any help would be greatly appreciated-I've been noodling around PS CS6 forever to no avail.

Not sure if this is what your looking for, but this is just a Gradient Map adjustment layer above the original. I added a pattern overlay layer style to generate the texture.

These are the colors of the gradient map sampled from your example.
